Background of Calponin 2
Calponin is a basic 34-kD protein first isolated from chicken gizzard and bovine aorta. It is specifically expressed in smooth muscle and binds calmodulin, actin, and tropomyosin. It is able to inhibit the ATPase activity of myosin and is though to play a role in smooth muscle contraction. Calponin is a smooth muscle specific, actin-, tropomyosin- and calmodulin-binding protein though to be involved in some way in the regulation or modulation of contraction.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: chicken heart tissue lysates,
Lane 2: chicken heart tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with mouse anti-Calponin/CNN2 antigen affinity purified monoclonal antibody (Catalog # MA1011) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-mouse IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1001) with Tanon 5200 system. A specific band was detected for Calponin/CNN2 at approximately 33 kDa. The expected band size for Calponin/CNN2 is at 33 kDa.

We are currently using anti-Calponin antibody (Monoclonal, CP-93) MA1011 for turkey tissue, and we are happy with the ICC results. The species of reactivity given in the datasheet says chicken, human, turkey. Is it possible that the antibody can work on horse tissues as well?

The anti-Calponin antibody (Monoclonal, CP-93) (MA1011) has not been tested for cross reactivity specifically with horse tissues, but there is a good chance of cross reactivity.
