Background of AGT
AGT (ANGIOTENSINOGEN), also called SERPINA8, is an alpha-2-globulin that is produced constitutively and released into the circulation mainly by the liver. ATG is a member of the serpin family, although it is not known to inhibit other enzymes, unlike most serpins. Angiotensinogen is also known as renin substrate. The AGT gene is mapped on 1q42.2. And the human angiotensinogen gene contains 5 exons. The expression of AGT and enzymes required for its conversion to angiotensin II in human adipose tissue. Western blot analysis showed a distinct band of expected size of the angiotensinogen protein (61 kD) in isolated adipocytes. The more pronounced AGT increase in TT subjects resulted in similar plasma renin activity at a lower plasma active renin concentration, with a higher plasma renin activity/active renin ratio.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: mouse liver tissue lysates,
Lane 2: mouse liver tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Angiotensinogen/AGT antigen affinity purified polyclonal antibody (Catalog # PA1643) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Angiotensinogen/AGT at approximately 55 kDa. The expected band size for Angiotensinogen/AGT is at 53 kDa.

Angiotensinogen/AGT was detected in a paraffin-embedded section of mouse liver tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-Angiotensinogen/AGT Antibody (PA1643) overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit (Catalog # SV0002) with DAB as the chromogen.

We are currently using anti-Angiotensinogen/AGT antibody PA1643 for mouse tissue, and we are happy with the WB results. The species of reactivity given in the datasheet says mouse, rat. Is it possible that the antibody can work on monkey tissues as well?

The anti-Angiotensinogen/AGT antibody (PA1643) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
