Background of ALK-1
Type I receptor for TGF-beta family ligands BMP9/GDF2 and BMP10 and important regulator of normal blood vessel development. On ligand binding, forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators. May bind activin as well.

We are currently using anti-ACVL1 antibody A01468-1 for rat tissue, and we are well pleased with the IHC results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on zebrafish tissues as well?

The anti-ACVL1 antibody (A01468-1) has not been validated for cross reactivity specifically with zebrafish tissues, though there is a good chance of cross reactivity. Would A01468-1 anti-ACVL1 antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?

Answer
You can see on the product datasheet, A01468-1 anti-ACVL1 antibody as been validated on WB. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
