Hedgehog Signaling Pathway

The Hedgehog signaling pathway is a key regulator of animal development and exists in all bilaterians. Members of the hedgehog family play key roles in a wide variety of developmental processes. Mammals have three known Hedgehog homologues: Desert (DHH), Indian (IHH), and Sonic (SHH). One of the best studied examples is the action of Sonic hedgehog during development of the vertebrate limb. Recent studies indicate the role of Hedgehog signaling in regulating adult stem cells involved in maintenance and regeneration of adult tissues. Disruption of hedgehog signaling during embryonic development can lead to severe developmental abnormalities. The pathway has also been implicated in the development of some cancers.

DNA Methylation and Transcriptional Repression Pathway

DNA methylation is the process by which methyl groups are added to the DNA molecule. This will change the activity of a DNA segment without changing the sequence. When located in a gene promoter, DNA methylation usually acts to repress gene transcription. DNA methylation and transcriptional repression are essential mechanisms to understand. They regulate functions such as cell differentiation, X-inactivation, gene expression, imprinting, and much more. Defects within the methylation machinery can result in genetic diseases such as Fragile X Syndrome, ICF, Rett Syndrome, and more. Transcriptional repressor proteins associate with target genes either directly through a DNA-binding domain or indirectly by interacting with other DNA-bound proteins.
