Research Spotlight - February 2023

In our February issue of Research Spotlight, we feature 10 recent publications from scientists whom Boster has served.

This month, several studies highlight the potential of nanotechnology innovations such as EPC-secreted exosomes, NIR-responsive hollow germanium nanospheres, berberin-enriched PLGA nanoparticles, and nanofiber-based meshes for improving the therapeutic output of nanomedicines.

Scientists also investigated how different groups of bioactive compounds, such as polyamine, vimentin, and dietary genistein exhibit immunomodulatory effects on the molecular pathways involved in the immune response against various disease conditions. In addition, novel roles for components of the SP3/PAD4 and miR-139-5p/ECE1 pathways in disease progression are discussed.

Manipulating Bioactivities of Endothelial Progenitor Cell-derived Exosomes for Promoting Angiogenesis in Ischemic Vascular Diseases

Authors: Dong, B., Qiu, Y., Liu, Z., Huang, J., Wang, Z., Tu, Q., Zhou, Z., He, J...
Journal: Nano Today

Exosomes paracrine-secreted by EPC (EPC-EXO) may exert strong pro-angiogenic effects, which offers a potential means to treat ischemic diseases. However, the mechanism of EPC-EXO-mediated angiogenesis remains unknown, and EPC-EXO shows poor homing to ischemic sites, which greatly limits the pro-angiogenic effect in vivo. We found...

Polyamine Metabolism Impacts T Cell Dysfunction in the Oral Mucosa of People Living with HIV

Authors: Mahalingam, S.S., Jayaraman, S., Bhaskaran, N., Schneider, E...
Journal: Nature Communications

Metabolic changes in immune cells contribute to both physiological and pathophysiological outcomes of immune reactions. Here, by comparing protein expression, transcriptome, and salivary metabolome profiles of uninfected and HIV+ individuals, we found perturbations of polyamine metabolism in the oral mucosa of HIV+ patients...

NIR-Responsive Hollow Germanium Nanospheres Mediate Photothermal/Photodynamic Therapy and Restrain Immunosuppression to Cooperatively Eradicate Primary and Metastatic Tumors

Authors: Li, Q., Fan, H., Xu, Y., Liu, M., Liu, J., Xu, L., Zou, M., Cheng, Q...
Journal: Chemical Engineering Journal

Photothermal and photodynamic therapies (PTT and PDT) generating regional hyperpyrexia or reactive oxygen species (ROS) not only kill tumors within irradiated site, but also potentially suppress metastatic tumors far away from irradiated site by activation of antitumor immunity through inducing immunogenic cell death (ICD) ...

Cholinergic Dysfunction-Induced Insufficient Activation of Alpha7 Nicotinic Acetylcholine Receptor Drives the Development of Rheumatoid Arthritis Through promoting Protein Citrullination via the SP3/PAD4 Pathway

Authors: Lv, C., Sun, M., Guo, Y., Xia, W., Qiao, S., Tao, Y., Fang, Y., Zhang, Q…
Journal: Acta Pharmaceutica Sinica B

Both cholinergic dysfunction and protein citrullination are the hallmarks of rheumatoid arthritis (RA), but the relationship between the two phenomena remains unclear. We explored whether and how cholinergic dysfunction accelerates protein citrullination and consequently drives the development of RA. Cholinergic function and protein...

Berberin Sustained-Release Nanoparticles Were Enriched in Infarcted rat Myocardium and Resolved Inflammation

Authors: Zhu, K., Yao, Y., Wang, K., Shao, F., Zhu, Z., Song, Y., Zhou, Z…
Journal: Journal of Nanobiotechnology

Inflammatory regulation induced by macrophage polarization is essential for cardiac repair after myocardial infarction (MI). Berberin (BBR) is an isoquinoline tetrasystemic alkaloid extracted from plants. This study analyzes the most likely mechanism of BBR in MI treatment determined via network pharmacology, showing that BBR acts...

Vimentin Epigenetic Deregulation in Bladder Cancer Associates With Acquisition of Invasive and Metastatic Phenotype Through Epithelial-to-Mesenchymal Transition

Authors: Reis, S.M., Gonçalves, V.M., Teixeira, C.G., Lima, C.M., Lobo, J…
Journal: International Journal of Biological Sciences

Bladder cancer (BlCa) is the ninth most common cancer worldwide, associated with significant morbidity and mortality. Thus, understand the biological mechanisms underlying tumour progression is of great clinical significance. Vimentin (VIM) is (over)expressed in several carcinomas, putatively in association with EMT. We have previously...

Dietary Genistein Increases Microbiota-Derived Short Chain Fatty Acid Levels, Modulates Homeostasis of the Aging Gut, and Extends Healthspan and Lifespan

Authors: Hou, Q., Huang, J., Zhao, L., Pan, X., Liao, C., Jiang, Q., Lei, J…
Journal: Pharmacological Research

Age-related gastrointestinal decline contributes to whole-organism frailty and mortality. Genistein is known to have beneficial effects on age-related diseases, but its precise role in homeostasis of the aging gut remains to be elucidated. Here, wild-type aging mice and Zmpste24-/- progeroid mice were used to investigate the role...

Authors: Liu, N., Zhou, Z., Ning, X., Zhang, X., Guo, Q., Guo, M., Wang, Y…
Journal: Materials & Design

The interaction between electrospinning nanomaterials and stem cells to repair damaged tissue mainly focus on manipulating cell viability, proliferation, or paracrine by the materials and their specific topology. In particular, the paracrine products of stem cells influence the behavior of those repairable cells (i.e., epidermal cells...
