Background of SOCS-4
SOCS4, also known as Suppressor of cytokine signaling 4, is mapped to 14q22.1. The protein encoded by this gene contains a SH2 domain and a SOCS BOX domain. The protein thus belongs to the suppressor of cytokine signaling (SOCS), also known as STAT-induced STAT inhibitor (SSI), protein family. SOCS family members are known to be cytokine-inducible negative regulators of cytokine signaling. Two alternatively spliced transcript variants encoding the same protein have been found for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: MCF-7 Whole Cell Lysate at 40ug,
Lane 2: SW620 Whole Cell Lysate at 40ug,
Lane 3: HELA Whole Cell Lysate at 40ug,
Lane 4: SKOV Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SOCS4 antigen affinity purified polyclonal antibody (Catalog # PB9506) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for SOCS4 at approximately 51 kDa. The expected band size for SOCS4 is at 51 kDa.

We are currently using anti-SOCS4 antibody PB9506 for human tissue, and we are well pleased with the WB results. The species of reactivity given in the datasheet says human. Is it likely that the antibody can work on monkey tissues as well?

The anti-SOCS4 antibody (PB9506) has not been tested for cross reactivity specifically with monkey tissues, though there is a good chance of cross reactivity.
