Background of GADD45 alpha
Growth arrest and DNA-damage-inducible protein GADD45 alpha (GADD45A) is a protein that in humans is encoded by the GADD45A gene. This gene is a member of a group of genes whose transcript levels are increased following stressful growth arrest conditions and treatment with DNA-damaging agents. It is located on 1p34-p12. Sequence analysis of human and hamster cDNA clones demonstrated that the gene has been highly conserved and encodes a novel 165-amino acid polypeptide. Northern blot analysis detected moderate expression of a 1.4-kb GADD45A transcript in heart, skeletal muscle, and kidney, with little or no expression in brain, placenta, lung, liver, and pancreas. In addition, Gadd45a promotes epigenetic gene activation by repair-mediated DNA demethylation.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: MCF-7 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GADD45A antigen affinity purified polyclonal antibody (Catalog # PB9945) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for GADD45A at approximately 19 kDa. The expected band size for GADD45A is at 18 kDa.

We are currently using anti-GADD45A antibody PB9945 for human tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human. Is it possible that the antibody can work on canine tissues as well?
