Background of FRA2
Fos-related antigen 2 is a protein that in humans is encoded by the FOSL2 gene. The Fos gene family consists of 4 members: FOS, FOSB, FOSL1, and FOSL2. These genes encode leucine zipper proteins that can dimerize with proteins of the JUN family, thereby forming the transcription factor complex AP-1. As such, the FOS proteins have been implicated as regulators of cell proliferation, differentiation, and transformation.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Rat Lung Tissue Lysate at 50ug,
Lane 2: HEPG2 Whole Cell Lysate at 40ug,
Lane 3: A549 Whole Cell Lysate at 40ug,
Lane 4: SW620 Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-FRA2 antigen affinity purified polyclonal antibody (Catalog # PB9591) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for FRA2 at approximately 43 kDa. The expected band size for FRA2 is at 35 kDa.

We are currently using anti-FRA2/FOSL2 antibody PB9591 for rat tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, rat. Is it true that the antibody can work on pig tissues as well?

The anti-FRA2/FOSL2 antibody (PB9591) has not been tested for cross reactivity specifically with pig tissues, though there is a good chance of cross reactivity.
