Background of EPCR
Endothelial protein C receptor (EPCR), also known as activated protein C receptor (APC receptor), is a protein that in humans is encoded by the PROCR gene. It has also recently been designated CD201 (cluster of differentiation 201). The protein encoded by this gene is a receptor for activated protein C, a serine protease activated by and involved in the blood coagulation pathway. And it is an N-glycosylated type I membrane protein that enhances the activation of protein C. Mutations in this gene have been associated with venous thromboembolism and myocardial infarction, as well as with late fetal loss during pregnancy. The encoded protein may also play a role in malarial infection and has been associated with cancer.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: A549 whole cell lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-EPCR/CD201 antigen affinity purified polyclonal antibody (Catalog # PB9880) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for EPCR/CD201 at approximately 46 kDa, 52 kDa. The expected band size for EPCR/CD201 is at 27 kDa.

We are currently using anti-EPCR/CD201/PROCR antibody PB9880 for human tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says human. Is it true that the antibody can work on canine tissues as well?
