Background of Carboxypeptidase A1/CPA1
Carboxypeptidase A1 is an enzyme that in humans is encoded by the CPA1 gene. This gene encodes a member of the carboxypeptidase A family of zinc metalloproteases. This enzyme is produced in the pancreas and preferentially cleaves C-terminal branched-chain and aromatic amino acids from dietary proteins. This gene and several family members are present in a gene cluster on chromosome 7. Mutations in this gene may be linked to chronic pancreatitis, while elevated protein levels may be associated with pancreatic cancer.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat pancreas tissue lysates,
Lane 2: mouse pancreas tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Carboxypeptidase A/CPA1 antigen affinity purified polyclonal antibody (Catalog # A05985-1) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Carboxypeptidase A/CPA1 at approximately 47 kDa. The expected band size for Carboxypeptidase A/CPA1 is at 47 kDa.

We are currently using anti-Carboxypeptidase A/CPA1 antibody A05985-1 for rat tissue, and we are content with the WB results. The species of reactivity given in the datasheet says human, mouse, rat. Is it likely that the antibody can work on monkey tissues as well?

The anti-Carboxypeptidase A/CPA1 antibody (A05985-1) has not been tested for cross reactivity specifically with monkey tissues, but there is a good chance of cross reactivity.
