Background of BMP-4
Bone morphogenetic protein 4 is a protein that in humans is encoded by BMP4 gene. It is found on chromosome 14q22-q23. The protein encoded by this gene is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. This particular family member plays an important role in the onset of endochondral bone formation in humans, and a reduction in expression has been associated with a variety of bone diseases, including the heritable disorder Fibrodysplasia Ossificans Progressiva. Alternative splicing in the 5' untranslated region of this gene has been described and three variants are described, all encoding an identical protein.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours.
Lane 1: Mouse Lung Tissue Lysate at 50ug,
Lane 2: Mouse Liver Tissue Lysate at 50ug,
Lane 3: HEPA Whole Cell Lysate at 40ug,
Lane 4: HEPG2 Whole Cell Lysate at 40ug,
Lane 5: HELA Whole Cell Lysate at 40ug.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-BMP-4 antigen affinity purified polyclonal antibody (Catalog # PB9688) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for BMP-4 at approximately 46 kDa. The expected band size for BMP-4 is at 46 kDa.

Question
We are currently using anti-BMP4 antibody PB9688 for mouse tissue, and we are satisfied with the WB results. The species of reactivity given in the datasheet says human, mouse. Is it likely that the antibody can work on canine tissues as well?

The anti-BMP4 antibody (PB9688) has not been tested for cross reactivity specifically with canine tissues, but there is a good chance of cross reactivity.
