Background of ICAM-2/CD102
Intercellular adhesion molecule 2 (ICAM2), also known as CD102 (Cluster of Differentiation 102), is a human gene. The protein encoded by this gene is a member of the intercellular adhesion molecule (ICAM) family. All ICAM proteins are type I transmembrane glycoproteins, contain 2-9 immunoglobulin-like C2-type domains, and bind to the leukocyte adhesion LFA-1 protein. This protein may play a role in lymphocyte recirculation by blocking LFA-1-dependent cell adhesion. It mediates adhesive interactions important for antigen-specific immune response, NK-cell mediated clearance, lymphocyte recirculation, and other cellular interactions important for immune response and surveillance. Several transcript variants encoding the same protein have been found for this gene.

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.
Lane 1: rat spleen tissue lysate,
Lane 2: rat thymus tissue lysate,
Lane 3: rat lung tissue lysate,
Lane 4: rat heart tissue lysate.
After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-ICAM2 antigen affinity purified polyclonal antibody (Catalog # A03965) at 0.5 μg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for ICAM2 at approximately 55KD. The expected band size for ICAM2 is at 31KD.

Can A03965 be used for neuroblastoma cells?

The Anti-ICAM2 Antibody Picoband™ (A03965) hasn't been used for neuroblastoma cells as our lab hasn't had the cells available in our sample panel. We suggest to run a pilot test to see if the antibody can work for the specific cells.

We are currently using anti-ICAM2 antibody A03965 for rat tissue, and we are happy with the ELISA results. The species of reactivity given in the datasheet says human, rat. Is it true that the antibody can work on feline tissues as well?
